The ultimate challenge of the Fortnite Chapter 2 "Open Water" Mission requests you to carefully analyse this week's loading screen. This is where you will find the clue that will lead you to the famous hidden 'O'.
As for the previous season, you will have to wait until you unlock the loading screen via temporary missions to hope to get your hands on the famous letter.

To find the hidden "O", you'll need to land in Craggy Cliffs. The secret letter is located below the building called "Sticks". You just have to destroy the ground to find the letter hidden underneath the building.
The Team Rumble mode is recommended to complete this challenge, since it allows you to come back to life quickly!
